At the International Symposium on Pediatric Neuro-Oncology (ISPNO), researchers presented findings from two trials aimed at overcoming resistance to standard BRAF/MEK-targeted therapies in patients with pediatric low-grade gliomas.
In this MedPage Today video, Lindsay Kilburn, MD, of Children's National Hospital in Washington, D.C., discusses the studies, which added hydroxychloroquine or everolimus (Afinitor) to targeted regimens in children with recurrent or refractory disease.

At ISPNO, I had the opportunity to present on two different clinical trials looking at combination therapy approaches for children with recurrent or refractory low-grade gliomas. And it's been really exciting over the last decades that BRAF alterations were identified as a really important molecular change in patients with low-grade glioma, and other molecular alterations, which activate that same kind of BRAF/MAP kinase pathway.
And that led to investigation of targeted therapies, MEK inhibitors and BRAF inhibitors, and really recently, very excitingly, FDA approvals of tovorafenib [Ojemda] and dabrafenib [Tafinlar]/trametinib [Mekinist] for treating children with pediatric low-grade glioma.
