Statin therapy may attenuate much of the atherogenic effects associated with leuprolide for treatment of prostate cancer, according to a post hoc analysis of the REVELUTION trial published in JACC: CardioOncology.
CVD is the leading cause of the death in men with prostate cancer, and adding statin therapy to leuprolide, a less expensive alternative to relugolix with a more favorable CV safety profile, may improve outcomes in this population, researchers reported.
“The REVELUTION ... trial recently demonstrated that the gonadotropin-releasing hormone agonist leuprolide, but not the gonadotropin-releasing hormone antagonist relugolix, was significantly associated with accelerated coronary plaque progression in men with localized prostate cancer,” Adithya K. Yadalam, MD, MSC, cardiology fellow at Emory University School of Medicine, and colleagues wrote. “Although relugolix may confer a more favorable cardiovascular profile, a nearly fivefold higher annual cost limits adoption.”
For the REVELUTION trial, researchers enrolled 62 men with nonmetastatic, treatment-naive prostate cancer and randomly assigned them to at least 6 months of leuprolide or relugolix (Orgovyx, Sumitomo Pharma/Pfizer) both on top of radiotherapy (mean age, 69 years; 56% on statin therapy).
